魟鱼软骨多糖对小鼠G422胶质瘤血管形成的影响
Effection of Ray Cartilage Glycosaminoglycans on G422 Glioma Angiogenesis
【摘要】 目的:观察魟鱼软骨多糖对小鼠G422胶质瘤血管形成及瘤组织VEGF mRNA和蛋白表达的影响。方法:建立小鼠G422胶质瘤腋下肿瘤模型,将实验分为生理盐水(Saline)组、不同剂量魟鱼软骨多糖(RCG)组、环磷酰胺(CTX)组,观察肿瘤生长情况,绘制肿瘤生长曲线,CD31免疫组织化学染色计数肿瘤组织微血管密度(M icrovessel density,MVD),采用RT-PCR和W estern b lot技术检测瘤组织血管内皮生长因子(Vascu lar endothelial growth factor,VEGF)mRNA和蛋白表达。结果:魟鱼软骨多糖各剂量组小鼠的肿瘤生长曲线平缓,MVD显著减少,VEGF mRNA和蛋白表达水平显著降低(P<0.01)。结论:魟鱼软骨多糖明显抑制小鼠G422胶质瘤的生长,抑制肿瘤的血管形成,可能与其下调肿瘤VEGF mRNA和蛋白表达有关。
【Abstract】 Objective:To explore the effection of Ray Cartilage Glycosaminoglycans(RCG)on the G422Glioma angiogenesis and VEGF mRNAand protein expressionin G422Glioma.Methods:The model of mouse with G422Gliomawas made,the experimental mice were randomly divided into normal saline group,RCG groups at varied concentrations and CTX group,Tumor growth states were observed;Microvessel density(MVD)was quantitated by immunohistochemistry using monoclonal antibodies of CD31;The expression of VEGF mRNAand protein in G422Gliomawas determined with RT-PCR and Western blot.Results :Tumor growth curves in RCG groups were smooth compared with Saline group;there were significant differences of MVD between RCG groups and Saline group;VEGF mRNA and proteinexpressionlevels in RCG groups were reducedsignificantly compared with Saline group.Conclusions:RCG could effectively inhibite the growth ofG422Gliomaandangiogenesis in G422Glioma,whichwas probably attributed to reducing theexpression ofVEGF mRNA and protein.
