小牛血去蛋白注射液对大鼠脑创伤后能量代谢的影响

【摘要】 目的研究小牛血去蛋白注射液(Actovegin,AC)预处理对于大鼠脑创伤(traumatic brain injury,TBI)后能量代谢的影响,探讨其对脑创伤保护作用的可能机制,从而为其临床应用提供实验依据。方法将40只大鼠随机分为4组,每组大鼠伤前腹腔注射AC或等容积生理盐水连续7天后制造大鼠脑创伤模型。伤后4小时每组各取10只大鼠采血,测定糖及乳酸含量。采血后将大鼠迅速断头,于伤侧大脑病灶周围取一块脑组织,测糖、乳酸、ATP、ADP及AMP含量并计算能荷(EC)。结果与假手术组相比,创伤组血糖含量明显升高,脑糖降低,血及脑中乳酸含量升高,脑组织ATP、ADP、AMP含量及EC下降。而AC逆转上述变化,且高剂量作用更加明显。结论AC对大鼠脑创伤有保护作用,且呈剂量反应关系,其作用是通过促进细胞能量代谢而实现的。

【Abstract】 Objective To investigate the impact of Actovegin(AC) protratement on energy metabolism in the rat brain following traumatic brain injury,so as to explore its possible functional mechanisms on TBI and offer foundations for its clinical application.Methods 40 SD rats were randomly divided into 4 groups.Traumatic brain injury models were made after sequent seven days’ peritoneal injection of Actovegin or normal saline.Blood was extracted through common carotid arteries of ten rats from each group 4 hours after injury and measurements of glucose and lactate content were made.Immediately after that,rats were decapitated and a piece of brain tissue was extracted surrounding the injury focus of each rat,glucose,lactate,ATP,ADP,AMP,and energy charge(EC) were calculated.Results Compared with the sham group,in the injury group the blood glucose value raised strikingly but glucose in the brain reduced,the content of lactate in both brain and blood arised prominently but ATP,ADP,AMP and EC went down strikingly.Actovegin turnovered the changes above and high dose produced more outstanding effects.Conclusions Actovegin can educe protective effects on rat TBI,and the effects are dose-related.Its probable mechanism is to facilitate energy metabolism.
