重组Tgo DNA聚合酶最适扩增条件的研究
Study on the Optimum Amplification Conditions for Recombined Tgo DNA Polymerase
【摘要】 以重组TgoDNA聚合酶为研究对象,检测了其最适扩增条件。结果表明:该重组酶的最适pH为8.75,最适Mg2+浓度为2.0 mmol/L,最适延伸温度为71.9℃。利用该重组酶成功扩增了质粒pUC19、野大麦Na+/H+逆向运转蛋白基因、拟南芥Timeless基因、小鼠Cx37基因和人CYT2C9基因。
【Abstract】 The optimum amplification conditions of the recombined Tgo DNA polymerase were examined in this study.The results revealed that the optimum PH of the enzyme was 8.75,the optimum concentration of Mg2+ was 2.0 mmol/L,and the optimum elongating temperature was 71.9 °C.Plasmid pUC19, Na+/H+ reversed transporting protein gene of wild barley,Arabidopsis Timeless gene,mouse Cx37 gene and human CYT2C9 gene were amplified successfully using this recombined enzyme.
