RP-HPLC分离纯化化学合成的催产素类似物

【摘要】 目的对化学合成的催产素类似物九肽进行分离纯化。方法采用RP-HPLC法。Develosil ODS C18柱,柱温为室温;流动相为15%乙腈+水+0.1%三氟乙酸;流速1 m l.m in-1;检测波长280 nm。结果进样量100 mg时分离效果最好,纯度可达96.5%。结论在最优化条件下,通过多次样品收集和冷冻干燥,可得到高纯度的催产素类似物。

【Abstract】 OBJECTIVE To purify a synthesized analogue of oxytocin,a nanopeptide in preparative scale.METHODS RP-HPLC method was adopted.Develosil ODS C18 column was taken,taking a mobile phase of 15% acetonitrile+water+0.1% TFA at the ambient temperature with a flow rate of 1.0 ml·min-1.The detection wavelength was 280 nm.Then the analytical methods were scaled up to preparative loading to obtain the nanopeptide.Different loading amounts were investigated for the effects on various parameters of preparative RP-HPLC.RESULTS The separate effect was excellent when the loading amount was 100 mg with the purity being 96.5%.CONCLUSION Under optimum separation conditions,the purified analogue of oxytocin was obtained through separation,collection and freeze-drying for many times.
