运动负荷对大鼠后肢骨成骨细胞整合素表达和血清骨钙素水平的影响
Effects of workload exerises on serum osteocalcin expression and integrins in the osteoblastso of SD rat’s femora
【摘要】 目的探讨运动负荷对SD大鼠成骨细胞整合素(integrin)的表达和血清骨钙素(BGP)水平的影响。方法挑选♀3周龄SD大鼠,运用高电压、低电流动物刺激仪,刺激SD大鼠进行主动跑跳训练,成功地模拟出运动负荷的动物模型。取鼠后肢骨分离的成骨细胞,分别在含10%胎牛血清的F-12培养基中培养、传代,再采用流式细胞技术测定和分析整合素α2、β1、β3在成骨细胞膜上的表达量;取鼠心脏血采用液体闪烁仪检测成骨细胞BGP的表达。结果运动负荷可显著改变SD大鼠成骨细胞整合素的表达及血清骨钙素的水平。结论运动负荷可直接对大鼠骨的形成产生有益的影响。
【Abstract】 OBJECTIVE To investigate that the effect of exercises on integrins and osteocalcin in the osteoblasts of Sprague-Dawley(SD) rats.METHODS 3-week-old female SD rats was chosed for high voltage and low current electrical stimulation,workload exercise model was successfully simulated.The osteoblasts isolated from SD rats femora were cultured in F-12 medium containing with 10% fetal bovine serum and subcultured.The expression of integrins α2,β1,β3 on the membrane of osteoblasts were studied with Flow Cytometry(FCM);BGP in blood from hearts were detected with LSC.RESULTS Exercises improve serum BGP and expressions of integrins α2,β1,β3 in the femoral osteoblasts of SD rat’s significantly.CONCLUSION Workload exercises could be beneficial to achieve bone remodeling.
