罗格列酮对大鼠胸主动脉损伤后TXB2和ET-1血浆水平的影响

【摘要】 目的通过观察罗格列酮对大鼠胸主动脉损伤后血栓素B2和内皮素-1血浆水平的影响,旨在探讨罗格列酮抗大鼠胸主动脉损伤后狭窄的机制。方法90只SD大鼠随机分为假手术组、损伤组和罗格列酮组。HE染色观察血管管腔面积和内膜面积变化;放免法测定血浆血栓素B2和内皮素-1的浓度。结果HE染色显示罗格列酮治疗后管腔面积较损伤组增加,内膜面积较损伤组减少(P<0.05);罗格列酮组血栓素B2和内皮素-1的血浆浓度较损伤组显著减少(P<0.05)。结论罗格列酮可以通过降低球囊损伤术后大鼠血栓素B2和内皮素-1血浆水平,减轻狭窄的发生。

【Abstract】 ObjectiveTo investigate the effects of rosiglitazone on the plasma levels of thromboxane B2(TXB2) and endothelin-1(ET-1) and explore its cellular mechanism in the inhibition of stenosis in thoracic aorta-injured rats. MethodsNinety SD rats were divided into three groups randomly: Sham group, Injury group and Rosiglitazone group. Thoracic aortas were isolated for observation of changes of the lumen and intima area under microscopy. The plasma levels of TXB2 and ET-1 was measured with radioimmunoassay. ResultsCompared with the injury group, the plasma concerntration of TXB2 and ET-1 was remarkably decreased, and the lumen area of thoracic aortas increased but the intima area of thoracic aortas remarkably decreased in the Rosiglitazone group. ConclusionRosiglitazone may decrease the release of TXB2 and ET-1,attenuate vascular stenosis in thoracic aorta-injured rats.
