黄芪注射液对兔钝性心肌损伤的影响

【摘要】 目的:探讨黄芪注射液(ASI)对兔钝性心肌损伤的保护作用。方法:健康中国家兔24只随机分为3组:对照组、实验组与ASI治疗组,其中实验组和ASI组利用BIMⅡ水平式生物撞击机撞击兔胸前区制备钝性心肌损伤模型。两组动物分别于撞击前5min,撞击后0.5,2,4,8及24h自股静脉采血检测cTnI、SOD、MDA。撞击后24h摘取心脏,制备超薄切片,透射电镜下观察其超微结构变化,对照组除不撞击胸部外,余处理均同实验组。结果:实验组及ASI组动物血浆中cTnI含量在撞击后224h均明显高于对照组(P<0.05),ASI治疗组升高幅度不如实验组,且较实验组相同时点有所下降(P<0.05);实验组SOD活性在撞击后0.5h即开始下降,伤后8h降低至最低点,伤后24h有所升高,但仍低于对照组及撞击前水平(P<0.01),ASI治疗组SOD活性与实验组同时点相比均有升高(P<0.05);实验组MDA含量自撞击后0.524h均高于对照组、撞击前水平(P<0.05),ASI治疗组MDA含量较实验组相应时点均有所下降(P<0.05);心肌超微结构观察治疗组损伤较轻。结论:黄芪注射液能减少家兔心肌损伤后血浆中cTnI及MDA水平,升高SOD的活性,对钝性心脏损伤具有一定的保护作用。

【Abstract】 Objective: To explore the effects of astragalus membranaceus injection(ASI) on myocardial blunt injury in rabbits. Methods: Twenty-four rabbits were randomly divided into the control group, experiment group and astragalus membranaceus injection treatment group (ASI group). BIM-ⅡHorizontal Bio-impact machine was used to make the model of myocardial blunt injury in the experiment and ASI group. Five minutes before and 0.5, 2, 4, 8 and 24 hours after impact, blood samples were taken from femoral vein of the animals to measure the concentration of serum myocardial troponin I (cTnI), the activity of serum superoxide dismutase (SOD) and the content of serum malondialdehyde (MDA). At the 24th hour after impact, the myocardial tissues were taken out and had electron microscope examination to observe morphological changes. Results: Compared with that of the control group, the concentration of serum cTnI during 2-24 hours post-impact and the content of serum MDA during 0.5-24 hours post-impact increased (P< 0.05), but the activity of serum SOD decreased during 0.5-24 hours post-impact(P<0.01) in the experiment group. Compared with that of the experiment group, levels of cTnI and MDA decreased (P< 0.05) and level of SOD increased (P<0.05) after impact in the ASI group. Morphological changes of myocardial tissues were remarkably ameliorated in the treatment group. Conclusion: Astragalus membranaceus injection can decrease the level of serum cTnI and MDA, and increase the activity of SOD after myocardial blunt injury in rabbits.
