有机铬对实验性糖尿病大鼠胰岛β细胞形态结构及功能的影响

【摘要】 目的探讨有机铬(2-吡啶甲酸铬)对大鼠胰岛β细胞形态结构及内分泌功能的影响。方法首先通过尾静脉注射新鲜配制的1.5%四氧嘧啶溶液制备糖尿病大鼠模型;通过灌胃的方式提高糖尿病大鼠体内有机铬含量;治疗12周后,通过氧化酶法测定大鼠血清葡萄糖水平,利用免疫组织化学方法和放射免疫学方法分别观察大鼠胰岛β细胞形态结构的改变及大鼠血清胰岛素含量的变化。结果两治疗组大鼠血清葡萄糖水平明显下降,与糖尿病模型组相比,差异均有显著性;400μg治疗组大鼠体内胰岛素水平明显升高,与糖尿病模型组相比差异有显著性,而200μg治疗组大鼠血清胰岛素水平虽有所增加但与糖尿病组相比,差异无显著性;免疫组化显示治疗组大鼠胰岛β细胞数量增多,胞质内胰岛素免疫反应阳性颗粒明显增多。结论有机铬对糖尿病大鼠具有明显的降低血糖功能,并能促进受损胰岛及β细胞形态结构功能的恢复,对糖尿病大鼠病理状态具有明显的改善作用。

【Abstract】 Objective To investigate the effect of organic chromium(2-pyridine chromic formate) on the structure and the endocrine function ofβ cells of rats by immunohistochemistry and radio-immunology.Methods The diabetes madel was established by injectingfresh 1.5% Alloxan from the caudal vein.The level of organic chromium was elevated by oral organic chromium liquid.After 12 weeks,the blood glucose was measuredby the oxidizing enzymemethod.The change in the morphologyof βcells in the isletsof the pancreasand in the level of blood serum insulin of rats were observed by immunohistochemistry and radio-immunology respectively.Results The blood glucosein the two treatment groups was obviously decreased compared with that of the diabetes group.The blood insulin of the 400μg-treated group was significantly increased compared with that of the diabetes group.The blood insulin of the 200μg-treated group was also increased,but showed no significant difference compared with that of the diabetes group.Immunohistochemistry showed an increase in the number of β cells and positivegranules in the cytoplasm of the treatment groups.Conclusion Organic chromium can effectivelydecrease the blood glucose and improvethe morphologand functionof βcells in the isletsof pancreas.It can obviously ameliorate the pathologic status of diabetic rats.
