花粉管通道法导入外源DNA技术及其在林木育种中的应用

【摘要】 本文综述了花粉管通道法导入外源DNA技术的创立、发展以及应用现状,指出花粉管通道法可以克服农杆菌介导、基因枪等遗传转化方法对组织培养的依赖,通过收获种子获得遗传多样性丰富的转化群体,选择余地大,可满足林木多无性系造林的要求;加之该方法与林木授粉后施加理化处理诱导三倍体技术同是操作雌花序,可以将两种育种技术联合使用,从而通过一轮次选育过程实现多目标性状遗传改良。随着研究的深入以及技术的日臻完善,花粉管通道法必将在林木育种领域发挥更大的作用。

【Abstract】 In the paper, the pollen-tube pathway were reviewed including its foundation, development and application situation. The ways of Agrobacterium inoculation and gene gun and so on heavily depended on tissue culture, while the pollen-tube pathway didn’t. Transformed clone from seeds with great genetic diversity which offer greater chance to meet the needs of forestation could be obtained via the pollen-tube pathway. In addition, both the pollen-tube pathway and female gamete chromosome doubling after pollination were operated on the female inflorescences. So they could be associated, and achieve multi-target traits genetic improvement only by one breeding process. As the research going on further and the technique developing, the method of pollen-tube pathway will play an important role in forestry breeding.
