脑膜瘤病理级别基因诊断芯片数学模型的建立

【摘要】 目的根据脑膜瘤基因表达谱,寻找脑膜瘤病理分级(WHO2000标准)的分子生物学依据,并建立脑膜瘤病理级别基因诊断芯片的数学模型。方法利用cDNA芯片技术建立24例脑膜瘤基因表达谱数据库,应用生物信息学分析软件GeneMaths2.0对脑膜瘤基因表达谱数据进行“非监督”样本聚类分析(Hierarchical聚类),将聚类结果与脑膜瘤样本的病理级别进行相关性分析;应用SAS9.0软件,用“贝叶斯判别法”建立脑膜瘤病理级别诊断的判别函数方程;对于脑膜瘤基因表达谱的数据可靠性,应用点杂交(DotBlot)实验进行了验证。结果脑膜瘤基因表达谱样本聚类结果与其样本的病理级别存在高度相关性;筛选出5条基因,建立了3个判别函数方程,根据它们的表达情况可以较准确的判断脑膜瘤的病理级别。结论脑膜瘤目前的病理分级标准有分子生物学基础;根据脑膜瘤基因表达谱可建立判断脑膜瘤病理分级的诊断芯片数学模型。

【Abstract】 Objective To establish the molecular classification and the diagnostic microarray for different pathological subtypes of meningiomas based on gene express pattern using cDNA microarray. Methods The total RNAs, extracted from 24 meningiomas and 3 normal brain tissues, were hybridized with the 14 112 genes fixed on cDNA microarrays (BioStarH140S) respectively, through which we obtained the 24 meningiomas gene express patterns. To analyze the gene express pattern, GeneMaths 2.0 and SAS 9.0 softwares were used to the Hierarchical clustering and Linear Discriminant Function establishment respectively. Further, the results obtained from the cDNA microarrays had been verificated by Dot Blot hybridization. Results The clustering results were highly correlated to the pathological subtypes of meningiomas. Five diagnosis genes were obtained and 3 Linear Discriminant Functions established, which could discriminate the histopathological subtypes of meningiomas accurately. Conclusion The histopathological subtypes of meningiomas have their molecular basis and the establishment of the diagnostic microarray will play an important role in elucidating the tumorgenesis and malignancy process in the future.
