WT1基因在白血病患者中的表达及临床意义

【摘要】 目的探讨WT1基因在白血病患者中的表达及其临床意义。方法运用RT-PCR方法检测WT1基因在50例白血病患者中的表达。结果急性髓细胞白血病(AML)和急性淋巴细胞白血病(ALL)WT1表达阳性率分别为77.3%(17/22)和54.5%(6/11)。慢性粒细胞白血病(CML)慢性期WT1表达均阴性,急变期55.6%(5/9)表达阳性。WT1阳性患者化疗缓解率低于阴性患者(分别为31.3%和69.2%,P<0.05)。结论WT1在白血病患者中高表达,可作为临床评估预后、检测微小残留病(MRD)的标志。

【Abstract】 Objective To evaluate the expression of WT1 gene in leukemia patients and its clinical implication. Methods Nest RT-PCR technique was applied to examine WT1 gene expression in 50 patients with leukemia. Results The WT1 mRNA positive rates in acute myeloid leukemias and lymphoblastic leukemias were 77.3% (17/22) and 54.5%(6/11) . In chronic myeloid leukemia, WT1 mRNA was undetectable at chronic phase ; but was revealed in 5 of 9 patients at blast crisis (55.6 %). The rate of remission in the patients positive for WT1 was lower than in those negative for WT1 (31.3 % vs 69.2 %, P <0.05). Conclusion The WT1 was highly expressed in the majority of the patients with leukemia and thus was involved in human leukemogenesis. It indicated WT1 could be a new prognostic factor and a panleukemia marker for the detection of MRD.
