【摘要】 目的:建立一种快速准确检验食品中单核细胞增生李斯特氏菌的方法。方法:采用聚合酶链反应 (PCR)特异性扩增单核细胞增生李斯特氏菌溶血素基因(hlyA),用PCR方法检测80份长春地区的食品样品。结果:在706 bp处出现hlyA基因的目的片段,食品样品中单核细胞增生李斯特氏菌阳性检出12份,阳性率为 15%,与吉林省疾病预防控制中心同步进行的国标法检测结果一致,二者符合率100%。结论:PCR方法检测食品中单核细胞增生李斯特氏菌更简便、快速、敏感、特异性高,适用于基层单位开展食品中单核细胞增生李斯特氏菌污染的调查及监测。

【Abstract】 Objective To establish a fast and accurate method to examine the Listeria monocytogenes in foods. Methods The polymerase chain reaction (PCR) method was adopted to amplify hlyA gene of Listeria monocytogenes. Eighty food samples of Changchun city were detected by PCR method. Results The target fragment of hlyA gene appeared at 706 bp. 12 positive results was gained in all detected samples, the positive rate was 15%. This result was identical with the detection result obtained by CDC of Jilin Province with international standard method, the coincidence rate was 100%. Conclusion PCR method is more concise, rapid, sensitive than other methods in detecting Listeria monocytogenes in food, the specificity is the highest. This method is applicable for basic sanitation supervision department to investigate and supervise the situation of Listeria monocytogenes contamination in food.
