银杏种仁总RNA提取方法

Method of the Isolation of Total RNA from the Ginkgo Biloba Seeds Contented Rich Polysaccharides and Secondary Metabolites

【摘要】 采用异硫氰酸胍法、TRIzol法、CTAB法和SDS法从成熟银杏种仁中提取总RNA,结果表明:CTAB法提取的总RNA纯度高,污染少,完整性好,其A260/A230为1.6~1.8,A260/A280为1.9~2.0;CTAB法提取的总RNA降解少,得率高;CTAB法更适于富含多糖和多酚物质的植物总RNA的提取。

【Abstract】 The isolation of high quality RNA from ginkgo biloba seeds is difficult due to high level of polysaccharides and other substances when the conventional procedures for RNA isolation were used.Four methods were used to isolate it and the results were compared.The CTAB procedure was the best.Following this procedure,we can obtaine good quality RNA with an average A260/A280 of 1.9~2.0 and A260/A230 of 1.6~1.8.The RNA was also examined with agarose gel electrophoresis.
