【摘要】 目的:建立反相HPLC法测定人血浆中利福喷丁的浓度。方法:以利福平为内标,采用ZORBAXSB-C18色谱柱(4.6mm×250mm,5μm),以甲醇-0.02mol·L-1磷酸缓冲液(67:33,v/v;pH7.0)为流动相,流速1.0ml·min-1,柱温40℃,340nm处紫外检测。结果:日内平均回收率为97.5%,RSD=4.1%(n=5),日间平均回收率为95.6%,RSD=5.4%(n=5),线性范围为0.54-17.28μg·ml-1(r=0.9995)。结论:本法简便快速,灵敏度高,重现性好,适用于利福喷丁化疗方案的临床监测及其药动学研究。更多还原

【Abstract】 Objective: To establish a RP-HPLC method for determination of rifapentine in human plasma. Method: Rifampin was used as the internal standard, the plasma samples were extracted. HPLC condition: analytical column; ZORBAX SB-C18(4.6×250 mm,5 μm) ,the mobile phase: methanol-0. 02 mol/L phosphate buffer (67: 33,V/V;pH7) ,the flow rate; 1.0 ml·min-1 ,the column temperature: 40 ℃,the detecting wavelength of UV:340 nm. Result: The chromatography was good and not interfered by the components of plasma. The intra-day average recovery was 97. 5% , RSD =4.1%(n=5) ,the inter-day average recovery was 95. 6% ,RSD = 5.4%(n=5), the linear range was within 0. 54-17. 28 μg·ml-1 ,r = 0.999 5. Conclusion: This is a simple, rapid, accurate, sensitive method for determination of rifapentine concentration.
