8-羟基脱氧鸟苷与吡啰红Y作用的共振光散射光谱及分析应用

【摘要】 目的:建立一种灵敏简便的样品中8-OH-dG分析新方法。方法:在pH 9.0的R.B缓冲液中,吡啰红Y阳离子染料与8-OH-dG阴离子形成离子缔合物,导致体系的共振光散射增强。结果:在优化的实验条件下,体系的共振光散射强度随着8-OH-dG的加入在339 nm处急剧增强。共振光散射的增强(ΔIRLS)与8-OH-dG加入量成线性关系,据此建立8-OH-dG的共振光散射检测新方法,方法的线性范围为50.0~226.4 ng/m l、检出限15.2 ng/m l。结论:该方法快速、简便,已成功用于加标样品的分析。

【Abstract】 Objective:To develop a simple and sensitive method for determination of 8-hydroxy-2′-deoxyguanosine (8-OH-dG).Methods:In the pH 9.0 buffer, the intensity of system′s resonance light scattering (RLS) was significantly enhanced due to the formation of ion-association complex of 8-hydroxy-2′-deoxyguanosine (8-OH-dG) anion with pyronine Y cation.Results:Under the optimal experimental conditions, a new RLS determination for 8-OH-dG was developed based on the enhancement(ΔIRLS) of the system′s RLS intensity at 339 nm, which was linear to the concentration of 8-OH-dG in the range 50.0226.4 ng/ml.The detection limit was 15.2 ng/ml.Conclusion:The proposed method succeeded in spiked sample determination with simple and sensitive characteristic.
