纤维蛋白原β基因148C/T及亚甲基四氢叶酸还原酶基因677C/T多态性与脑梗死的关系研究
A Statistical Study on the Relationship between Gene Polymorphisms of β-fibrinogen 148C/T,N5,N10-methylene-tetra-hydrofolic Acid Reductase 677C/T and Cerebral Infarction
【摘要】 目的探讨纤维蛋白原(Fg)β基因148(β148)C/T及亚甲基四氢叶酸还原酶基因(MTHFR)677C/T多态性与脑梗死易感性的关系。方法按年龄、性别、有无高血压病史及糖尿病史选取相匹配的病例组及对照组各100例,利用聚合酶链反应-变性高效液相色谱法(PCR-DHPLC)确定Fgβ148及MTHFR 677基因型。结果大动脉粥样硬化组(TOAST分型)及相应对照组Fgβ148 CC、CT/TT基因型分布差异有显著性(P=0.035),余各组两个多态位点的基因型分布差异无显著性。吸烟或饮酒的FgβCT/TT基因型携带者在病例组和对照组的分布差异有显著性(均P<0.05)。结论Fgβ148 CT/TT基因型可能是大动脉粥样硬化性脑梗死的危险因素,携带FgβCT/TT基因型同时吸烟或饮酒可能是脑梗死的危险因素。
【Abstract】 Objective To study the relationship between gene polymorphisms of β-fibrinogen(Fgβ)148C/T,N5,N10-methylene-tetra-hydrofolic acid reductase(MTHFR) 677C/T and cerebral infarction.Methods 100 patients with cerebral infarction and 100 controls matched for age(within 3 years),race,sex,history of hypertension and diabetes mellitus(yes or no) were enrolled in the study.Genotyping for Fg 148C/T and MTHFR 677C/T gene polymorphism was performed by polymerase chain reaction and the technique of denaturing high performance liquid chromatography(PCR-DHPLC).Results There was statistically significant difference of the Fg 148 CC,CT/TT genotype distribution between large-vessel subtype group(TOAST criteria) and correspond-ing control group(P=0.035).The differences of distribution of Fgβ148 CT/TT genotype in combination with smoking or drinking between cases and controls were statistically significant.Conclusions The Fgβ148 CT/TT genotype might be genetic risk factor for large-vessel subtype group of cerebral infarction.Fg 148 CT/TT(genotype) combined with smoking or drinking might be susceptible predictor for cerebral infarction.
