小鼠局灶性脑缺血的偏瘫症状检测
Evaluation of the Tests for Neurological Injures Induced by Focal Cerebral Ischemia in Mice
【摘要】 目的 探索小鼠局灶性脑缺血后偏瘫症状检测的方法。方法 采用大脑中动脉阻塞法(MCAO)造成小鼠持续性局灶性脑缺血,缺血后24 h通过悬挂试验、爬板试验综合评分法和Bederson症状评分法检测局灶性脑缺血小鼠的偏瘫症状;后处死动物取脑,切成1 mm厚冠状脑片,在5 g/L氯化三苯四氮唑(TTC)溶液中孵育 30min,将TTC染色脑片图像输入计算机,以AnalyPower1. 0软件计算每一脑片的梗死体积;将前囟-2 .0 mm处脑片切成8μm厚冷冻切片并进行苏木精 伊红(HE)染色,于高倍光镜下计算神经元密度。结果 悬挂、爬板试验综合评分法测得的偏瘫症状与TTC染色法测得的大脑梗死体积有较好的相关性(Spearman等级相关分析,rs=0 .713,P<0. 001);前者与传统的Bederson症状评分法评定的偏瘫症状进行比较结果较为一致(r=0 .631,P<0. 001)。结论 用悬挂、爬板试验综合评分法可客观、定量评估脑缺血诱导的偏瘫程度,并与神经损伤程度及大脑梗死灶大小基本一致。
【Abstract】 Objective To explore the method that estimate neurological function deficit of focal cerebral ischemia in mice. Methods Persistent focal cerebral ischemia was induced by middle cerebral artery occlusion (MCAO) in mice using intraluminal suture. The neurological function deficit was quantificationally evaluated by integrated grades of hanging test and climbing board test 24 h after the surgery and was followed by Bederson mothed. The brains were then removed and coronally dissected into 1mm thick sections. The sections were stained with 0.5 g/L TTC at 37 ℃for 30 min and the images were recorded and stored in the computer. The volumes of infarcts by the TTC staining were calculated using an image analyzer(AnalyPower10). Coronary setions(8 μm) were prepared from fifth brain slice (bregma-2.0 mm), and stained with HE, and the neurons density es was obtained by light microscopy examination. Results A close correlation existed between integrated grad es(hanging test and climbing board test) and the volumes of infarcts by the TTC staining (r s=0713,P<0001). The integrated gradesof hanging test and climbing board test were correlated closely to Bederson scores (r=0631,P<0001) Conclusions The integrated grade of hanging test and climbing board test is the objective index of neurological function deficit of focal cerebral ischemia that is consistent with the volumes of infarcts in the focal cerebral ischemic mice.
