致病疫霉(Phytophthorainfestans)的分离与培养方法

【摘要】 针对不同地区的病样,分别采用直接分离法、保湿培养分离法、薯片夹叶分离法、薯片诱导分离法等分离方法,利用选择性培养基SRA进行分离,通过挑取菌落边缘菌丝尖端于斜面培养基进行纯化,共获得重庆马铃薯和番茄主产区186个致病疫霉菌株。测定代表性菌株在不同培养基上的菌落生长和产孢量。结果表明,致病疫霉在RSA上生长最快,RA和RSB次之;在RSB上产孢最多,RTA次之。

【Abstract】 Routine isolation way, dew chamber way, disease leaf claping with potato slices and induction with potato tuber were tested, besed on semi-composed medium of RSA plus several antibiotics. Total 186 isolates from representive areas of potato and tomato in Chongqing were obtained and purified through cutting the top of mycelium of pathogen. Bioassay of typical isolate was carried out to determine the growth and sporulation of Phytophthora infestans. The results showed that the growth rate of Phytophthora infestans on tested media is as following order: RSA>RA>RSB>SVA>RTA>LVA>VA>MA>PSA>PDA; RSB is best for sporulation, then RTA.
