几种植物雌激素对细胞间隙连接通讯的影响

【摘要】 目的 了解植物雌激素对细胞的细胞间隙连接通讯 (GJIC)的影响。方法 用荧光漂白后恢复技术,在激光扫描共聚焦显微镜下观察典型的植物雌激素 (槲皮素、染料木黄酮、拟雌内醇和肠内脂)对正常人皮肤角质形成细胞株 (HaCaT细胞 )GJIC的影响。结果 不同种类的植物雌激素在不同浓度下对GJIC的影响不完全相同。槲皮素在 0 .1 ~10. 0μmol/L浓度下对HaCaT细胞的GJIC功能无明显影响。染料木黄酮、拟雌内醇和肠内脂在 0. 1~10. 0μmol/L浓度下,能不同程度地抑制GJIC功能,荧光恢复率为 31 .77% ~37.. 06%,显著低于溶剂对照组 (44 74% )。结论 除黄酮类槲皮素外,常见的植物雌激素 (染料木黄酮、拟雌内醇和肠内脂 )在人体可能达到的血清浓度下,对HaCaT细胞的GJIC功能都有不同程度的抑制,提示在一定条件下可能有促癌作用。因此,将其用作药品或保健食品时应该慎重。

【Abstract】 Objective To observe effect s of phytoestrogens quercetin (QC),Genistein (GEN), coumestrol (COM), and enterol actone (ENL) on gap junctional intercellular communication (GJIC) in HaCaT cells .Methods HaCaT cells were exposed to QC, GEN, COM,and EN L at 0.1, 1.0, 10.0 and 100.0 μmol/L for 24 hours.The effects of phytoest rogens on GJIC were determined by fluorescence redistribution after photobleachi ng (FRAP) technique of using a laser scanning confocal microscope (LSCM). Results QC did not affect the GJIC at 0.1-10.0 μmol/L, whe reas, GEN, COM, and ENL exhibited inhibition on the GJIC in some extent at 0.1 -10.0 μmol/L without showing significant cytotoxicity.The ratio of fluoresc ence recovery were between 31.77% to 37.06%, which were significantly decrease d compared the vehicle control (44.74%).Conclusion The phytoestrogens GEN, COM, and ENL, but not QC, could inhibit the GJIC founction i n HaCaT cells at concentrations could be reached in human serum in some instance , indicating they could, under certain conditions, be cancer promoters.Therefo re, it should be prudent to use these chemicals as pharmaceuticals or dietary su pplements.
