Nm23-H1基因对膀胱癌化疗敏感性的体外实验研究

【摘要】 目的: 探讨外源性人抑癌基因nm23 H1 对膀胱癌细胞系T -24 细胞化疗敏感性的影响。方法: 将nm23 -H1真核表达质粒通过脂质体转染技术导入人膀胱癌细胞株T -24,利用免疫组化及Western blot法鉴定质粒的表达。利用MTT法、流式细胞仪分析技术检测T -24细胞转染前后化疗药物对细胞增殖、细胞凋亡的变化。结果: 顺铂组(CDDP)细胞生长明显受到抑制,细胞凋亡增加;顺铂转染前后细胞凋亡指数分别为(26.51±0.98)%、(63.21±0.58)%,两者间差异有统计学意义。结论: Nm23- H1 基因能增强顺铂的化疗敏感性,为基因联合化疗药物治疗膀胱癌提供了参考依据。

【Abstract】 Objective: To evaluate the effect of exogenous human tumor suppressor gene nm23-H1 on the chemotherapeutic sensitivity of the bladder cancer cell lines T-24 cell. Methods: the plasmid of nm23-H1 was transferred into human bladder cancer cell lines T-24 cell by the method of lipofect transfermation and the expression of nm23-H1 were assayed by means of immunohistochemical technique and Western blot MTT assay was used to assess the cell proliferation effect of the chemotherapeutic medicines on the bladder cancer cell lines T-24 cell transferred by nm23-H1, and the cell apoptosis was measured by flow cytometry. Result: The grows of T-24 cell with cisplatin(CDDP), transferred by nm23-H1, obviously was subjected to be repressed,and the cell apoptosis was to run away to add;and the indexes of the cell apoptosis of two groups, transferred and not transferred by nm23-H1, were separately (26.51±0.98)%、(63.21± 0.18)%;Two groups have a significant difference. Conclusion: Gene of nm23-H1 can enhance the chemotherapeutic sensitivity with cisplatin(CDDP), and supply the reference of the gene-chemical therapy for the bladder cancer.
