抗寒霜减轻大鼠冻区组织损伤的能力及其机制

【摘要】 目的:通过预防性用药,观察大鼠在寒冷条件下冻区组织形态学的病理变化,评价抗寒霜对减轻组织损伤,提高御寒能力的作用,为寒区部队预防冻伤提供有效的药物及给药途径。方法:制备大鼠左后足冻伤模型。实验分3组,对照组为冻伤模型组,实验1组为抗寒Ⅰ霜组,实验2组为抗寒Ⅱ霜组,全麻下取大鼠冻区组织,观察其病理改变。结果:3组大鼠冻区的表皮组织、外周神经纤维、肌肉组织、血管组织均有明显的病理改变,其中对照组与实验2组大鼠的表皮组织均有明显的空泡样变性其发生率明显高于实验1组(P<0.01),3组大鼠均有不同程度的血管内损伤,管腔内有大量的红细胞堆积,其中对照组与实验2组的发生率明显高于实验1组(P<0.05),对照组与实验2组均有肌纤维断裂或肌浆红染现象,表明肌肉组织的代谢发生障碍。实验结果还表明,3组大鼠的周围神经纤维髓鞘均有退行性变。结论:抗寒霜对提高寒冷条件下大鼠的御寒能力,减轻组织损伤具有一定的作用。

【Abstract】 AIM:To observe the rats’ pathological changes in histomorphology under cold condition,comment the use of anti cold cream for reducing tissue damage and improve ability of resisting cold by means of preventive medication so as to provide the army in cold area with effective drugs and way of administration to prevent cold injury.METHODS:Cold injury models of rats’ left hind feet were prepared.The rats in this experiment was divided into three groups:the group of cold injury models as control group,experimental group 1 treated with anti cold cream Ⅰ and experimental group 2 with anti cold cream Ⅱ .The tissues in cold area were taken under general anaesthesia to observe its pathological changes.RESULTS:Great pathological changes were found in epidermal tissues,peripheroneural tissues,muscular tissues and vascular tissues in cold area of all rats,among which,the rats’ epidermal tissues in the control group and the experimental group 2 had obvious vacuole like denaturation and their occurrence rate was higher than that in the experimental group 1(P< 0.01).All rats had various damage in the blood vessels.There was a great quantity of red blood cell(RBC) accumulation in the vascular cavity,and the occurrence rate of RBC accumulation in the control group and the experimental group 2 was obviously higher than that in the experimental group 1(P< 0.05).The rats had muscular tissue crack or reddening staining appearance in the muscular cytoplasma in the control group and the experimental group 2,showing the obstruction of metabolism in the muscular tissues.The experimental result also indicated that there were degradation changes in the myelin sheath of the peripheral nerve fiber of all rats.CONCLUSION:Anti cold cream has certain effects in improving the rats’ ability to resisting cold under cold condition and reducing tissue injury.
