大鼠外周血淋巴细胞钟相关基因的筛选和鉴定
Screening and Identification of Clock-related Genes in Peripheral Lymphocytes of Rat
【摘要】 目的探讨外周血淋巴细胞的钟相关基因。方法采用mRNA差异显示技术比较不同时点基因的差异显示,筛选出大鼠外周血淋巴细胞的钟相关基因,并进一步用RNA干涉技术确定其为钟下游基因。结果获得10条差异表达的cDNA片段,其中3种与已知基因过氧化氢酶、髓鞘蛋白脂蛋白和组蛋白乙酰化酶同源,4种与已知EST同源,另有3种为未知EST,它们均属Clock基因的下游基因。结论外周血淋巴细胞中至少存在3种以上钟相关基因,其表达水平接受作为昼夜节律振荡器重要组分的Clock基因的调控。
【Abstract】 Objective To explore clock-related genes in peripheral blood lymphocytes of rats.Methods We used the method of differentially displayed polymerase chain reaction(DD-PCR) to display the differential genes at two timepoints,and then applied the technique of RNA interference to confirm the relationship between the screened EST and the Clock gene.Results Ten differential(cDNA) fragments were found,among which there were 3 known genes such as catalase,myelin(proteolipid) protein and histone acetylase,4 known EST and 3 novel EST.The levels of expression is all these screened genes or EST were regulated by the Clock gene.Conclusion There are at least more than three clock-related genes in peripheral lymphocytes of rat and their expressions is regulated by the Clock gene as an important component of the circadian oscillator.
