雄性核不育水稻矮秆突变体突变分子机制的初步研究
A Preliminary Study on the Mutational Mechanism of Genic Male-sterile Rice Dw arfing Mutants
【摘要】 以株-1S、SV1S、SV14S为研究材料,分析这3种材料的胚乳α-淀粉酶活性、第二叶鞘及节间长度对赤霉素(GA3)反应,结果表明来源于株-1S的SV1S、SV14S突变体的矮化变异与赤霉素信号传导途径无明显关系.此外,通过3种材料的基因组DNA和线粒体DNA的RAPD分析,发现矮秆突变体株系的基因组DNA和线粒体DNA都存在一定的变化,从而在DNA分子水平上进一步证实了SV1S和SV14S突变体的遗传突变.其突变的生理机制与赤霉素信号传导途径无明显关系,很可能与赤霉素生物合成途径有关.
【Abstract】 Zhu 1S and the dwarfing mutants SV1S and SV14S were used as the pl an t materials and the response of the activity of α amylase of the endosperm,the second leaf sheath and internodes to exogenous GA3 were analyzed.The results sh owed that the dwarfing variation of SV1S and SV14S derived from Zhu 1S were not due to the gibberellin signal transduction.The RAPD analysis of the genomic DNA and mitochondria DNA of Zhu 1S,SV1S and SV14S showed that there were signifcan t variations in the genomic DNA and mitochondrial DNA of the dwarfing mutants,wh ich further comfirmed the genetic mutations of SV1S and SV14S at the DNA molecul ar level.The cause of the dwarfing phenotype was suggested not due to the gibber ellin signal transduction rather being be associated with the gibberellin biosyn thesis.
