人胚神经干细胞体外长期培养系统的建立
Establishment of long-term human neural stem cell culture system
【摘要】 目的 建立人胚神经干细胞的体外长期培养系统并研究其生物学特性。方法 从胚龄为2 0周的人胚胎皮质组织分离出神经干细胞 ,连续传代培养并检测生长曲线 ,测定长期培养对细胞周期和细胞分化及冻存的影响。结果 体外培养神经干细胞达到了 10个月 ,传代 2 5代 ,总的细胞数增加了 10 5倍。干细胞只有培养 1个月后才能有效去除前体细胞。细胞周期显示细胞保持了活跃的增殖 ,多次传代后细胞冻存对细胞的存活力无明显影响 ,多向分化潜力表现出稳定性。结论 人神经干细胞可于体外长期培养 ,这也是细胞纯化的有效方法。
【Abstract】 Objective To set up the long-term culture system of human neural stem cells (hNSCs) in vitro and study the biological properties. Methods Isolating hNSCs from human fetus of about 20 weeks, growt h curve of continually passaged cells was examined, then the effect of long-ter m culture on cell cycle, cell differentiation and cryopreservation were analyzed .Results hNSCs culture had been expanded up to 10 5 fold, and passaged for 25 times in 10 months. Only one month after primary culture pr ecursors could be effectively discarded. Cell cycle analysis indicated that hNSCs remained remarkable proliferation. Cell survival was not affected by cell cryopreservation after different passages. Multilineage potential of differe ntiation remained unchanged. ConclusionhNSCs can be cultured in vitro for long time which can be thought as an effective method to purify hNS Cs.
