化纤方对肺纤维化大鼠之肺系数及肺组织形态学影响的实验研究
Experimental Study on Effects of “Hua Xian Fang” on Pulmonary Coefficients and Morphological Changes In Rats of Pulmonary Fibrosis
【摘要】 目的观察化纤方对博莱霉素所致肺纤维化大鼠肺组织形态学的影响,探索其阻抑肺纤维化的效应与机制。方法健康SD大鼠,随机分为6组:空白组、模型组、化纤方高剂量组、化纤方中剂量组、化纤方低剂量组、泼尼松组。气管内注入博莱霉素制作大鼠肺纤维化模型,造模后7d开始治疗,化纤方分别按成人剂量的20倍、10倍、5倍灌胃,连续治疗30d后处死动物,观察各组治疗后肺组织病理学变化以及比较各组肺系数。结果化纤方各组及泼尼松组肺系数值明显降低(P<0.01),肺组织病理学显示肺泡炎及肺纤维化程度均明显减轻。结论化纤方具有一定的阻抑肺纤维化作用。
【Abstract】 Objective To investigate the effects of “Hua Xian Fang” on pulmonary fibrosis and pulmonary coefficients in rats induced by bleomycin and explore its mechanism. Methods Healthy SD rats were randomly allocated into 6 groups:normal group,model group,the high-dosage of“Hua Xian Fang” group,the middle-dosage of“Hua Xian Fang” group,the low-dosage of “Hua Xian Fang” group,and the prednisone group. The six groups except for the normal group were injected with bleomycin via trachea to induce experimental pulmonary fibrosis, and lavage respectively with “Hua Xian Fang” at 20 times, 10 times, 5 times of adult-dosage on the 7th day after medication. After a month, all rats were sacrificed and the pulmonary pathological changes and pulmonary coefficient were observed in each group. Results The pulmonary coefficients of the every “Hua Xian Fang” group and the prednisone group compared with the model group were obviously reduced (P< 0.01). The histopathological examination markedly reduced alveolitis and pulmonary fibrosis. Conclusion It is revealed that “Hua Xian Fang” has some effects on bleomycin-induced pulmonary fibrosis and smaller side-effect than Prednisone.
