应用反向间接血凝试验检测山羊痘病毒抗原
The detection of goat pox virus antigen by reverse passive hemagglutination
【摘要】 以兔抗山羊痘病毒IgG致敏绵羊红细胞,采用反向间接血凝试验(RPHA)检测山羊痘病毒抗原。该诊断试剂与山羊痘病毒抗原发生特异性的凝集反应;在PBS中不自凝;与其它动物病病毒抗原无交叉反应。对贵州省 8个疫区现场采集山羊痘待检抗原检测结果表明,该方法阳性检出率为 87. 5% ( 7 /8 ),与琼脂扩散试验(AGP)阳性检出率为 50%(4 /8)相比较,RPHA敏感性比AGP高;同时对山羊痘野生强毒H-GZ株F1 ~F5 代细胞培养物进行检测,F1 代就可检出RPHA效价,而AGP则不能检测。该诊断试剂可用于兽医临床上山羊痘的快速诊断及作为病毒分离培养的检测指标。
【Abstract】 The sheep erythrocyte was sensitized with IgG purified from rabbit anti-goat pox virus serum and reverse passive hemagglutination(RPHA) has been developed in this experiment. The diagnostic reagent could specifically reacted with goat pox virus antigen , no self-agglutination in phosphorous buffer solution(PBS) and no cross reaction with other animal virus antigens were observed. The samples were collected from affected goats in 8 areas of Guizhou province. The results indicated that the positive rate of RPHA reached 87.5%(7/8),as compared with the positive rate of 50%(4/8) in agar gel precipitation(AGP),RPHA was more sensitive than AGP. Meanwhile detection was conducted with F1-F5 cell cultures infected by virulent virus H-GZ strain, the virus antigen could be detected in RPHA since F1 generation and no reaction with all cell cultures in AGP. The reagent could be used for diagnosing goat pox quickly and working as an indicator for virus isolation and cultivation.
