【摘要】 目的研究胃癌中Survivin基因的表达情况及其生物学意义。方法选择胃癌42例和正常胃黏膜24例,对Survivin、Bcl-2和CyclinD1抗体行SP法检测。结果42例胃癌标本中Survivin有35例阳性,其中28例在胞质内表达,7例在核内。24例正常胃黏膜组织中有5例阳性表达(P<0.001)。CyclinD1和Bcl-2在胃癌中的阳性率分别为47.6%(20/42),33.3%(14/42)。Survivin和CyclinD1表达未显示相关性(P=0.062),与Bcl-2呈负相关(P=0.031)。Survivin基因表达均与性别、部位、淋巴结转移无相关性。但在高、中分化胃癌中的强阳性(++～+++)表达率明显高于低分化腺癌(P=0.005)。结论Survivin基因蛋白既可在胃癌中表达,又可在正常胃黏膜中表达;除表达于细胞质外,也可表达于细胞核。Survivin独立于Bcl-2之外在肿瘤的发生、发展中起重要作用。更多还原

【Abstract】 Objective To study the biological significance of the expression of Survivin in gastric cancer patients and its correlation with CyclinD1 and Bcl-2. Methods 42 cases of gastric carcinomas and 24 cases with normal gastric mucosas were detected for CyclinD1, Bcl-2 and Survivin with immunohistochemistry. Results In 42 cases of gastric carcinomas, 35 were Survivin positive. Among them, 28 were expressed in tumor cell cytoplasm while 7 in nuclei. 5 out of 24 cases with normal gastric mucosas were Survivin positive (P < 0.01). The positive rates of CyclinD1 and Bcl-2 in gastric carcinomas were 47.6%(20/42),33.3%(14/42), respectively. Significant inverse relationship was found between Survivin and Bcl-2(P=0.031). Survivin expression was not correlated with clinical pathological parameters such as sex, site and lymph node metastasis. Nevertheless, we found the strong expression(++～+++)rate of high and mediate differentiated cases was significantly higher than that of low and un-differentiated cases(P=0.005). Conclusions The expression of Survivin gene may be correlated with gastric carcinomas, showing a higher positive rate in gastric carcinomas (83.3%) than in normal cases (20.8%). Survivin is independent from Bcl-2 on the development of gastric carcinomas.更多还原
