不同人群TTV感染的检测及部分基因序列分析

【摘要】 目的探讨湛江地区不同人群中TTV的感染状况及部分TTV的基因序列。方法应用PCR检测512份正常献血员血清标本、60份HBsAg阳性血清标本和48份抗-HCV阳性血清标本的TTV感染状况,并对部分扩增出的阳性片段进行克隆测序,与国内外报道的序列进行同源性比较。结果正常献血员、HBsAg阳性、抗-HCV阳性标本的TTVDNA阳性率分别为10.94%、13.33%和16.67%,不同人群间结果无显著性差异(P>0.05);随机选出的5例TTVDNA阳性片段,其序列显示与日本株(AB008394)和中国株(TTVCH1)相应位置核苷酸序列的同源性大于97%,可能属同一基因型。结论湛江地区不同人群TTVDNA与HBsAg阳性、抗-HCV阳性无明显相关。

【Abstract】 Objective To investigate status of TTV infection and analysis of the gene sequence from different groups in Zhanjiang City. Methods The 512 samples from blood donors, 60 from HBsAg positives and 48 from anti-HCV positives were collected and detected for the infectious status of TTV, and some of the sequence of positive fragments amplified were analyzed by using PCR. Results The positive rates of TTV DNA in these three groups were 10.94%,13.33% and 16.67%, respectively. There were no significant difference among the three groups (P>0.05). The sequence analysis showed that there were above 97% nucleotide identity among AB008394 (from Japan),TTV CH1(from China) and our 5 isolates. Conclusion The positive rate of TTV DNA was not obvious correlated with the positive rates of HBsAg and anti-HCV in different groups of Zhanjiang City.
