川芎嗪抑制Ⅱ型胶原诱发关节炎大鼠滑膜损伤作用机制的研究
Mechanism of inhibition effect of ligustrazine on synovium lesion in rats with arthritis induced by collagen II
【摘要】 目的观察川芎嗪对大鼠胶原性关节炎(CIA)炎症发展和滑膜分泌血管内皮生长因子(VEGF)的影响。方法皮内注射Ⅱ型胶原诱导大鼠CIA,给予不同剂量川芎嗪灌胃,以环孢霉素A(CSA)为阳性对照。检测关节炎指数积分和炎症足垫厚度,X摄片检测关节结构,透射电镜观察滑膜毛细血管内皮细胞超微结构,免疫组化观察滑膜VEGF的表达和滑膜微血管密度(MVD)。结果与模型组比较,CSA和100mg/(kg·d)川芎嗪可降低关节炎指数积分和炎症足垫厚度,减轻炎症关节结构的破坏,改善滑膜毛细血管内皮细胞超微结构,抑制滑膜表达VEGF和减少滑膜MVD。结论川芎嗪可抑制CIA大鼠关节炎症发展和关节结构的破坏,其机制与川芎嗪可抑制滑膜表达VEGF、从而抑制滑膜组织血管生成密切相关。
【Abstract】 Objective To observe the influences of ligustrazine on the development of rat collagen induced arthritis(CIA) and vascular endothelial growth factor(VEGF).Methods Rats were given intradermal injection of Collagen II to induce CIA,and then different doses of ligustrazine was perfused,and to take cyclosporin A(CSA) as a positive control.The index score and corn thickness of arthritis were detected and joint structure was examined with X ray.The synovial hemoendothelial ultrastructure was observed by transmission electron microscope(TEM)and the expression of synovial VEGF and microvessel density(MVD) were observed by the immunohistochemistry.Result Compared with the model group CSA and 100 mg/kg.d of ligustrazine decreased the index score and corn thickness of arthritis,relieved the lesion of joint structure,improved the synovial hemoendothelial ultrastructure,inhibited the expression of VEGF and reduced MVD of synovium.Conclusion Ligustrazine can inhibit the development of arthritis and lesion of joint structure in CIA rats.The mechanism may be related to the inhibition effect on the expression of VEGF and angiogenesis in synovium.
