异丙酚对肺缺血再灌注大鼠中性粒细胞的影响
Effects of propofol on polymorphonuclear leukocytes in lung ischemia-reperfusion in rats
【摘要】 目的 探讨静脉麻醉药异丙酚对肺缺血/再灌注(I/R)大鼠中性粒细胞(PMN)的影响。方法 夹闭大鼠左肺门4 5min ,再灌注2h ,建立在体肺I/R模型。大鼠随机分为3组:假手术组、缺血再灌注组、异丙酚组(I/R并用异丙酚维持麻醉)。用流式细胞仪测定全血PMNCD18的表达和呼吸爆发(RB) ,测定左肺顺应性和湿干比(W /D) ,测定血浆MDA和SOD含量。结果 麻醉剂量的异丙酚抑制PMNCD18的表达和RB ;改善再灌注后左肺顺应性和降低W/D ;降低血浆MDA水平并保护SOD的活力。结论 麻醉剂量的异丙酚对肺I/R大鼠全血PMN具有抑制作用,对肺I/R损伤具有保护作用。
【Abstract】 Objective To investigate the effects of propofol on the polymorphonuclear leukocytes(PMNs) in the lung ischemia-reperfusion(I/R) in rats.Methods A rat model of pulmonary warm I/R injury was established by clampping of the left lung hilus for 45min and then reperfusion for 2h.All rats were divided into 3 groups:sham (without clampping),I/R and propofol groups (I/R with propofol anesthesia).We measured the CD18 expression and respiratory burst (RB) of PMNs by flow cytometry,the compliance and wet to dry (W/D) weight ratio of the left lung,the levels of MDA and SOD in plasma.Results In pulmonary I/R injury,propofol at anesthetic dose inhibited CD18 expression and RB of PMNs,increased the compliance and decrease W/D of the left lung,reduced MDA levels,preserved SOD activity.Conclusion Propofol at anesthetic dose inhibit the activity of neutrophils and have protective effects on the pulmonary I/R injury in rats.
