氯化D,L-肉碱腈的制备和拆分研究

【摘要】 对用氯化3 氯 2 羟丙基三甲胺(CHPTMA)制备氯化D,L 肉碱腈的工艺条件进行了研究,最佳的反应条件是:反应温度40~45℃,反应时间1.5h,物料CHPTMA与氰化钠的摩尔比为1∶1.05。在此条件下,产率为78.8%。研究了利用N 乙酰 L 谷氨酸为拆分试剂对氯化D,L 肉碱腈进行拆分,氯化L 肉碱腈的一次拆分光学纯度为92%,产率为37%;二次拆分光学纯度达95%。

【Abstract】 The preparation of D,L-cyano-carnitine chloride from 3-chloro-2-hydroxypropyl trime-thylamine chloride was studied. The best reaction conditions are:reaction temperature is 40~45℃; reaction time is 1.5 h; molar ratio of 3-chloro-2-hydroxypropyl trimethylamine chloride and sodium cyanide is 1∶1.05. Under the conditions, the yield of D,L-cyano-carnitine chloride is 78.8%. The separation of the enantiomers of D,L-cyano-carnitine chloride was also studied using N-acetyl-L-glutamic acid as the resolution agent. The enantiomer purity of L-cyano-carnitine chloride after the first resolution is 92% with 37% yield. The Enantiomer purity after the second resolution is 95%.
