蛋白质聚集体对膜污染过程的影响

【摘要】 BSA在溶液中存在少量二聚体,这对BSA的吸附和膜污染过程有很大影响.实验结果显示,BSA可以通过分子间二硫键在膜表面形成聚集体.通过加入还原性试剂DTT(二硫苏糖醇)可减少二聚体的形成,从而达到降低膜污染的目的.通过丙烯酸改性膜分离过程,说明静电作用也是BSA污染过程中的重要因素.这些结果有助于阐明蛋白质对膜的污染机理,为解决膜污染问题提供新的思路.

【Abstract】 Bovine serum albumin (BSA) forms dimer or oligomer in it s aqueous solution, which has a significant effect on its chromatographic behavi or and the adsorption of BSA on membrane surface.The experimental results obtain ed by the present study demonstrated the formation of BSA aggregate via inte rmolecular disulfide bonds. Addition of low concentration of DTT (dithiothreitol )effectively reduced the formation of BSA aggregate, and thus enhanced the filtr ation process. Modification of membrane by acrylic acid showed that electrostat ic interaction also played an important role in the membrane fouling caused by B SA adsorption.The results described above provided important insights into the p hysical and chemical interactions governing membrane fouling by protein adsorpti on, which might inspire new approaches to reducing membrane fouling.
