反相高效液相色谱法快速测定长春花中4种生物碱
Rapid Determination of 4 Vinca Alkaloids by Reversed Phase High Performance Liquid Chromatography
【摘要】 采用反相高效液相色谱法同时分离测定 4种长春花吲哚类生物碱。以水∶二乙胺 =986∶14,用磷酸调节pH=7. 2(溶液A),甲醇∶乙腈 =4∶1(溶液B), 380mLA和 620mLB混合作为流动相,流速 2mL/min,进样量 10μL, 220nm下检测,以恒流洗脱方式在 25min内分离了长春碱等 4种成分。同时还研究了流动相组成、pH值等对分离效果的影响。对实际样品分析,取得了较好结果。
【Abstract】 A method for the rapid determination of 4 vinca alkaloids by reversed phase high performance liquid chromatography (RP HPLC) was proposed. These 4 ingredients were separated on a Diamonsil (C 18 column 250 mm×4.6 mm i.d., 5 μm ) using water∶diethylamine=986∶14 adjust pH to 7.2 H 3PO 4, as solvent A and methanol∶ acetonitrile=4∶1 as solvent B, and the mobile phase was composed of 380 mL A and 620 mL B. The detection was performed at a flow rate of 2.0 mL/min, and detected at 220 nm within 25 min. The effect of distribution proportion of the mobile phase and pH value were discussed. And from the sample analysis we get the optimal result.
