青紫薯色素对β淀粉样肽诱导神经细胞毒性的作用
EFFECT OF SWEET-POTATO ANTHOC YANINS ON CYTOTOXICITY OF PC-12 CELL INDUCED BY AMYLOID β PROTEIN
【摘要】 ①目的探讨青紫薯色素对β淀粉样肽诱导神经细胞毒性的保护作用及其机制。②方法建立β淀粉样肽诱导的PC-12神经细胞毒性病理模型,MTT法测定青紫薯色素对PC-12神经细胞增殖活性的影响,流式细胞仪检测PC-12神经细胞的凋亡率、线粒体膜电位、细胞内Ca2+浓度和Caspase-3的活性。③结果青紫薯色素可以剂量依赖性地提高PC-12细胞的增殖活性,降低β淀粉样肽诱导的PC-12细胞凋亡率,减轻线粒体膜的损伤,降低细胞内钙离子的浓度,抑制Caspase-3的活化。④结论青紫薯色素通过抑制Caspase-3的活化,减轻线粒体膜的损伤,降低细胞内Ca2+的浓度,起到减轻β淀粉样肽诱导的PC-12细胞损伤的作用。
【Abstract】 Objective To investigate the effect of sweet-potato anthocyanins on cytotoxicity of PC-12 cells induced by amyloid β(Aβ) protein. Methods A pathological model of PC-12 cell cytotoxicity induced by Aβ was established. MTT was applied to measure the effect of sweet-potato anthocyanins on PC-12 cell proliferation ; flow cytometry for analysis of the cell viability, apoptosis, activity of caspase-3, and mitochondria membrane potential, and intracellular Ca2+ in PC-12 cells. Results The anthocyanins can dose-dependently increase the PC-12 cell viability, decrease the apoptosis of the cells, improve the mitochondria membrane potential,and decrease the concentration of intracellular Ca2+, and inhibit the activation of caspase-3. Conclusion The protective effect of sweet-potato anthocyanins on PC-12 cell injury induced by Aβ can be reached by ways of inhibition of caspase-3 activation, improvement of mitochondria membrane potential, and a decrease of the concentration of intracellular Ca2+.
