药物电针刀疗法对兔膝骨性关节炎软骨细胞凋亡的影响
Effect of Drug-Electric Acupotomy Therapy on Rabbit Articular Chondrocyte Apoptosis
【摘要】 目的研究药物电针刀疗法对骨关节炎家兔一氧化氮(NO)及软骨细胞凋亡的影响。方法应用关节制动的方法复制骨关节炎(OA)模型,18只造模成功的中国家兔,随机分为药物电针刀组(简称 A组)、液针刀组(简称 B 组)和对照组(简称 C 组),A 组用药物电针刀加手法治疗,B 组用液针刀加手法治疗,C组仅予以手法治疗,每3日1次,疗程为1个月。用硝酸还原法检测关节滑液中 NO 含量;用流式细胞仪检测软骨细胞的凋亡率。结果 A、B、C 组关节滑液中 NO 的含量分别为(33.67±17.72)μmol·L-1、(58.90±22.07)μmol·L-1和(83.30±16.20)μmol·L-1,软骨细胞的凋亡率分别为(25.27±10.82)%、(51.71±12.42)%和(74.80±9.37)%,3组间差异有显著性。结论与液针刀加手法相比较,药物电针刀疗法加手法能更显著减少关节液 NO 的含量,抑制软骨细胞的凋亡,而治疗膝骨性关节炎。
【Abstract】 Objective To study the effects of drug-electric acupotomy(DEA)therapy on nitric oxide(NO)content and articu- lar chondrocyte apoptosis in rabbit with osteoarthritis(OA).Methods The rabbit OA model was established by immobi- lized in full extension for six weeks.Eighteen successfully duplicated OA model rabbits were randomly divided into the DEA group(treated with DEA and manipulation),the acupotomy group(treated acupotomy and manipulation)and the control group(treated with manipulation alone).The treatment was applied once every three days for 1 month as a course.NO content in articular synovia was measured by nitrate reduction method,and the apoptosis rate of chondrocyte was determined by flow cytometry.Results NO content in the A,B,and C groups was 33.67±17.72 μmol/L, 58.90±22.07 μmol/L and 83.30±16.20 μmol/L respectively.The apoptosis rate in the threc groups was 25.27± 10.82%,51.71±12.42% and 74.80±9.37% respectively.Comparison among the three groups showed statistical sig- nificant difference.Conclusion Compared with the acupotomy plus manipulation,DEA plus manipulation could reduce the NO content in articular synovia and inhibit the apoptosis rate of chondrocytes more effectively in treating osteoarthritis of knee joint.
