【摘要】 目的观察中药补肾活血方治疗免疫介导型再生障碍性贫血小鼠的疗效,并探讨其作用机制。方法建立免疫介导型再生障碍性贫血动物模型,观察中药补肾活血方的疗效,并从造血祖细胞集落培养和骨髓基质细胞黏附功能,造血调控因子等方面探讨其作用机制。结果中药补肾活血方可改善免疫介导型再障小鼠的外周血象,增加骨髓有核细胞计数,延长其生存期,促进红系造血祖细胞 CFU-E、BFU-E 的增殖、分化,调节 T 细胞亚群,抑制 Ts 的活性,促进白细胞介素-10(IL-10)的分泌,抑制造血负调控因子白细胞介素-2(IL-2)、干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)的活性。结论中药补肾活血方通过促进 CFU-E、BFU-E的增殖、分化以及调节细胞免疫功能而达到治疗的目的。更多还原

【Abstract】 Objective To observe the effect of Bushen Huoxue recipe(BHR)in treating immune mediated aplastic anemia mice and to explore its mechanism.Methods The animal model of immune mediated aplastic anemia was established to observe the effect of BHR on it,and the therapeutic mechanism was explored viewing from the aspects of hemopoietic stem/ancestor culture,adhesive function of bone marrow stromal cells and hemopoietic regulatory factors.Results BHR could improve the peripheral blood figure in the model mice,increase the number and prolong the lifespan of nucleated cells in bone marrow,promote the proliferation and differentiation of CFU-E and BFU-E,regulate T-lymphocyte subsets, inhibit the activity of Ts cells,advance the secretion of IL-10,and suppress the activity of the negative hemopoietic regu- latory factors such as 1L-2,IFN-7 and TNF-α.Conclusion BHR is effective in treating aplastic anemia by way of pro- moting proliferation and differentiation of CFU-E,BFU-E,and regulating cellular immune function.更多还原
