海湾扇贝面盘幼虫“面盘解体病”病原的研究

【摘要】 对大连地区患"面盘解体病"的海湾扇贝(Argopectenirradians)面盘幼虫进行电镜观察,并对健康海湾扇贝幼虫进行人工感染实验。患该病面盘幼虫病症为:下沉,组织解离,游泳器官面盘解体,经过一段时间后全部死亡。对发病面盘幼虫进行超薄切片电镜观察发现,该病原病毒主要侵染面盘幼虫的结缔组织细胞和上皮细胞,病毒呈六角形或近圆形,未成熟病毒颗粒直径为84~90nm,病毒颗粒形态不一,大致有4种与所报道的鸡马立克氏病毒(Marek’sdiseaseherpesvirus)和火鸡疱疹病毒(Turkeyherpesvirus)相似,即致密拟核型、中空型、十字型和双环型。从形态学上进行鉴定,认为该病毒属于疱疹样病毒(Similarherpesvirus)。人工感染实验显示,在感染病毒后100h,有30%幼虫出现"面盘解体"病状;感染148h,幼虫全部死亡(水温22~24℃)。根据实验结果推断,病毒的传播途径包括垂直传递和水平传播。

【Abstract】 An investigation was conducted along the sea area of Dalian, Liaoning Province. The Argopecten irradians veligers with ’facial disc disintegration disease’ were obsevered under electron microscope and the artificial infection experiment was made. The symptoms showed that the diseased veligers subsided and their tissues dissolved with the facial disc of swimming organ disintegrating, and all died at last. The pathogenic virus mainly invaded the connect tissue cell and epithelial cell of the veliger. The virus had the shape of hexogan or approximate spherical, and the diameter of immature virions were 84-90 nm. The virions were various in shape, and four morphological types were similar to marek’s disease herpes virus and turkey herpes virus, which were compact nucleoid type, hollowing type, crisscross type and two-ring type. The artificial infecting results show that 30% veligers appeared the symptom of ’facial disc disintegration ’ after 100 h infection, and all died after 148 h at water temperature 22-24 ℃. The transmission ways of the virus includes vertical and horizontal transmission.
