编码TCR可变区的DNA免疫诱导BALBC小鼠调节性免疫应答
DNA vaccination encoding variable region gene of the T cell receptor induces regulatory immune response in BALB/C mice
【摘要】 目的 :研究编码卵清白蛋白 (OVA)T细胞克隆TCRVα5 2或Vβ2 1的DNA免疫正常小鼠诱发调节性免疫应答。方法 :用重组质粒pcDNA3 1 Vα5 2或pcDNA3 1 Vβ2 1免疫BALB C小鼠 ,RT PCR证实重组质粒能在体内和体外真核细胞中转录。3H TdR掺入法分析细胞增殖 ,3H TdR标记的靶细胞检测杀伤T细胞的杀伤效应 ,免疫荧光法分析血清中特异抗体水平和Vβ2 + T细胞数量。结果 :pcDNA3 1 Vα5 2或pcDNA3 1 Vβ2 1能在体内和体外真核细胞中转录 ,重组质粒免疫BALB C小鼠能诱导产生特异性体液免疫应答、T细胞增殖反应及对靶细胞CTL反应 ,但pcDNA3 1 Vβ2 1免疫没有导致Vβ2 + T克隆清除 ,而是细胞活性部分封闭。结论 :编码TCRVα5 2或Vβ2 1的DNA免疫能诱导正常BALB C小鼠调节性免疫应答。
【Abstract】 Objective:To investigate regulatory immune response induced by DNA vaccination encoding T cell receptor Vα5.2 or Vβ2.1 chain predominantly displayed on ovalbumin (OVA)-specific T cell clone.Methods:BALB/C mice were vaccinated with pcDNA3.1 encoding T cell receptor (TCR) Vα5.2 or Vβ2.1 chain respectively.Using RT-PCR,transcription of the recombined plasmids was analysed. Cell proliferation was measured by 3H-TdR incorporation.CTL response was assayed by JAM test.Immuno-fluorescent assay was used to examine the anti-TCR antibody level and the number Vβ2 +T cells.Results:RT-PCR analysis showed that the recombined plasmids can be transcripted in vivo and in vitro.DNA vaccine with TCR variable chain effectively induced TCR-specific humoral and cellular immune response,Vβ2 +T cells was not depleted by Vβ2.1 TCR-DNA vaccination,but rather was anergy.Conclusion:Regulatory immune response can be induced by DNA vaccination encoding TCR Vα or Vβ region in normal mice.
