【摘要】 目的:探求ERCC2表达与人卵巢癌实体瘤发生、耐药的关系。方法:分别取23例卵巢癌实体瘤、癌旁正常组织、培养及加药培养卵巢癌实体瘤组织各100 mg,提取组织中总RNA,采用RT-PCR的方法分析细胞中ERCC2表达水平。结果:5例标本中,ERCC2在卵巢癌实体瘤表达显著低于癌旁正常组织。20例实体瘤标本经更生霉素、环磷酰胺、顺铂、5-氟尿嘧啶、阿霉素处理后ERCC2分别增加18.2 %、72.0 %、104.9 %、14.1 %、23.2 %。结论:ERCC2在部分卵巢癌实体瘤患者中低表达,可能是这些卵巢癌实体瘤患者细胞恶变形成的基础。卵巢癌实体瘤患者对更生霉素、环磷酰胺、顺铂、5-氟尿嘧啶、阿霉素耐药程度不同。更多还原

【Abstract】 Objective:To search the association between expression of ERCC2and drug resistance in human ovarian tumor.Methods:Total ribonucleic acid was extracted from 23 patients with ovarian tumor and adjacent normal tissues cultured with KSM;CTX;DDP;5-Fu;ADM or not,quantitative analysis ERCC2 gene expression by reverse transcription-polymerase chain reaction. Results:In5patients,ERCC2gene ex-pression in ovarian tumor was evidently lower than that in adjacent normal tissues.The expression levels of ERCC2in20ovarian tumor culture with KSM;CTX;DDP;5-Fu;ADM increased18.2%;72.0%;104.9%;14.1%;23.2% compared with those without that.Conclusion:ERCC2gene expression in ovarian tumor was lower than in adjacent normal tissues,and may be the cause of canceration in partial ovarian tumor.KSM;CTX;DDP;5-Fu and ADM had a different effect on ERCC2expression in ovarian tumor and drug resistance.更多还原
