改良凝集试验用于东方田鼠血清中弓形虫抗体的检测

【摘要】 目的 探讨弓形虫改良凝集试验 (MAT)检测野生东方田鼠 (Microtusfortis)血清中弓形虫抗体的特异性。 方法 用MAT测定野生东方田鼠血清中弓形虫抗体 ,同时对实验室内繁育并感染过血吸虫尾蚴的东方田鼠血清进行检测。 结果 检测 12 4份洞庭湖野生东方田鼠血清 ,弓形虫抗体阳性 3 6份 ,阳性率为 2 9.0 % ,阳性血清滴度为 1∶2 0者 11份(8.9% )、1∶40者 4份 (3 .2 % )、1∶80者 4份 (3 .2 % )、1∶160者 2份 (1.6% )、1∶3 2 0者 2份 (1.6% )、1∶64 0者 2份 (1.6% )、1∶12 80者 2份 (1.6% )、1∶2 5 60者 1份 (0 .8% )、>1∶2 5 60者 8份 (6.5 % )。 10 4只实验室繁育并且感染过日本血吸虫尾蚴的东方田鼠血清没有检测到弓形虫特异的抗体。 结论 野生东方田鼠弓形虫感染较普遍 ,抗体水平高 ;MAT抗原与血吸虫感染的动物宿主血清无交叉反应 ,表明MAT具有高度的特异性。

【Abstract】 Objective To investigate specificity of modified agglutination test (MAT) detecting antibodies to Toxoplasma gondii in serum of field mice Microtus forti. Methods Serum of trapped field mice were detected for T. gondii specific antibodies with the MAT using formalin-fixed tachyzoites,and serum of laboratory-bred field mice infected with Schistosoma japonicum were also detected. The serum were diluted two-fold starting at 1∶20 in pH 7.2 phosphate-buffered saline containing 0.2 mol/L 2-mercaptoethanol on 96 well microtiter plate. The dilution of 1∶20 serum was as the threshold titre for detecting antibodies to T. gondii. Results Thirty-six (29%) out of 124 trapped field mice was found to be positive for antibodies to T. gondii. The antibody titers of positive sera (% in parentheses) were 1∶20 (8.9%),1∶40 (3.2%),1∶80 (3.2%),1∶160 (1.6%),1∶320 (1.6%),1∶640 ( 1.6 %),1∶1 280 (1.6%),1∶2 560 (0.8%),and >1∶2 560 (6.5%). No antibodies to T. gondii were detected in 104 sera of laboratory-bred field mice even they were infected with S. japonicum between 1 and 45 days post infection. Conclusion T. gondii infection is more prevalent in wild field mice M. forti. Nonspecific reactions of the MAT antigen were not found in sera from laboratory-bred field mice infected with S. japonicum,indicating it is highly specific for detecting T. gondii infection.
