改良反相高效液相色谱法测定非清髓性异基因干细胞移植患者全血中环孢素A的C2浓度

【摘要】 目的 :建立了一种简单的环孢素A(CsA)的萃取方法和反相高液相色谱测定法。方法 :全血乙醚 石油醚 (7∶1 )单步提取后 ,经正己烷脱脂后进样 ;以C1 8为固定相 ,凹形梯度洗脱 ,(A液 :乙腈 水 (44∶1 1 ,内含 0 .0 2 %TFA) ,B液 :乙腈 ,1 2min内从A变化到B ,凹形梯度 ) ,检测波长 2 1 4nm ,柱温 5 5~ 70℃。结果 :CsA在 0 .0 5~ 2 0mg·L-1 范围内呈良好的线性关系。结论 :此方法提取步骤少 ,方法简单快速、专一性强、费用低 ,适合临床CsA的血药浓度监测 ,并测定了 1例NHLⅢB期患者非清髓性异基因干细胞移植后早期全血C2 浓度。

【Abstract】 OBJECTIVE A new simple extraction of cyclosporin A in whole blood and an improved method of HPLC were developed. METHODS CsA was extracted from the whole blood by ethyl ether: petroleum ether(4∶1),degreased by hexane, then analyzed in C 18 column by concave gradient (solution A: acetonitrile-water(44∶11) with 0.02% TFA; solution B: aetonitrile, A was transformed to B within 12 min by concave gradient).CsA was detected in 214 nm with CsD as internal standard. The column temperature was 55~70℃. RESULTS The method is linear within the range of 0.05~20 mg·L -1.CONCLUSION The method is simple, rapid, sensitive and applied to determine the therapeutic drug monitoring. The C 2 in a NHL ⅢB patient after nonmyeloablative alleogentic peripheral blood stem cell transplantation was determined.
