MMP-2抑制剂对成釉细胞瘤细胞体外侵袭的影响

【摘要】 目的:探讨基质金属蛋白酶-2抑制剂Ro31-9790对成釉细胞瘤细胞体外侵袭的影响。方法:通过给予Ro31-9790对体外培养的成釉细胞瘤细胞进行处理,采用明胶酶谱法、流式细胞术、体外侵袭试验、细胞粘附分析、免疫组化、MTT试验等方法,观察Ro31-9790对成釉细胞瘤细胞体外侵袭等特性的影响。结果:Ro31-9790对成釉细胞瘤细胞的侵袭及粘附能力均有明显的抑制作用;4例成釉细胞瘤细胞均表达MMP-2,但不同肿瘤组织来源的成釉细胞瘤细胞MMP-2表达量不同。Ro31-9790能显著抑制成釉细胞瘤细胞分泌的MMP-2的活性,但不影响成釉细胞瘤细胞MMP-2和TIMP-2的表达;Ro31-9790不能抑制成釉细胞瘤细胞的体外增殖。结论:MMP-2的活性及其相关的细胞粘附能力与成釉细胞瘤的侵袭能力密切相关,Ro31-9790可在体外抑制成釉细胞瘤的侵袭

【Abstract】 PURPOSE: To investigate the effects of matrix metalloproteinase-2(MMP-2)inhibitor Ro31-9790 on the invasion of ameloblastoma cells and the mechanisms in vitro. METHODS: Ameloblastoma cells were treated with Ro31-9790 and the effects of Ro31-9790 on the invasion of ameloblastoma cells were observed. Gelatin zymography, flow cytometry, in vitro invasion assay, cell attachment assay, immunohistochemistry and MTT assay were employed to study the mechanisms of the effects produced by Ro31-9790. RESULTS: Ro31-9790 could inhibit the invasion and attachment and MMP-2 activity of ameloblastoma cells in vitro. In ameloblastoma cells of 4 patients, positive expression of MMP-2 was found, but was different in the cells from various patients. Ro31-9790 did not inhibit the expression of MMP-2 and TIMP-2 and the proliferation of ameloblastoma cells in vitro. CONCLUSION: MMP-2 activity can contribute to the invasive capacity of ameloblastoma. Ro31-9790 can inhibit the invasion of ameloblastoma cells in vitro.
