新型多聚体超声造影剂的初步实验研究
Preliminary Experimental Study on an Intravenous Polymer Echo Contrast Agent
【摘要】 目的 观察自制新型多聚体超声造影剂的基本特性及其对实验犬左心室及心肌增强显像的效果。方法 扫描电镜观察微泡大小形态及粒径分布 ,Coulter计数仪测定微泡浓度 ,4只杂种犬其中 2只制成前壁心肌梗死模型 ,经股静脉 0 .5 ml团注新型多聚体超声造影剂观察犬左室及心肌显影效果 ,同时监测心率及呼吸频率。结果 该新型多聚体造影剂为含空气的冻干粉剂 ,平均粒径 1.5μm,10 0 %的微泡粒径小于 8μm,通过周围静脉注射使左心室腔产生明显的增强显像 ,其中左心室 2 +以上程度的显影占总注射次数的 92 % ,肺转运时间为 2~ 3个心动周期。左心室显影峰值出现在左心室显影后 3~ 5个心动周期 ,持续高强度显影 15 s以上 ,触发成像模式可见心肌显像同时能检测梗死区心肌。造影过程中未见明显的心率、呼吸及心电图改变。结论 新型多聚体超声造影剂可以获得良好的左心室及心肌增强显像 ,对实验犬的心率、呼吸无明显影响。
【Abstract】 Objective:To evaluate the basic characteristics and effects of a novel polymer echo contrast agent on the left ventricular cavity and myocardial opacification with experimental animal study.Methods:Scanning electron micrographs and Coulter counter analysis were done to determine the bubble size,morphology and concentration.Four dogs including 2 acute myocardial infarction models were studied to evaluate the acoustic effect of the polymer echo contrast agent.Left ventricular and myocardial echo images were recorded before and after intravenous bolus injecting 0.5 ml of the agent with ultrasound machine.Results:The novel polymer air agent was formulated as a lyophilized product,with a number of mean diameter of 1.5 μm and 100% of the bubbles was below 8 μm. Satisfactory echo contrast images of the left ventricular cavity and myocardium were got after intravenous injection of the agent.The transpulmonary time was about 2~3 cardiac cycles,the left ventricular (LV) high intensity opacificaton lasted more than 15 seconds and 92% of intravenous injections showed perfect LV opacification.The heart and respiratory rate as well as electrocardiogram monitoring did not show any abnormal changes during the study.Conclusions:The result of this study suggests that this novel polymer echo contrast agent be effective and safe in left ventricular and myocardial echo contrast test with high quality of the left ventricular cavity and myocardium contrast images.
