【摘要】 目的:探讨三苯氧胺(tamoxifen,TAM)和甲孕酮(medroxyprogesterone,MPA)联合用药对顺铂耐药的卵巢癌细胞株SKOV-3／DDP生长影响的机制。方法:用四甲基偶氮唑蓝(MTT)法、电子显微镜及流式细胞仪测定用药前后SKOV-3／DDP细胞生长、形态学、周期及凋亡率的变化。结果:低浓度的TAM和MPA联合用药组对SKOV-3／DDP细胞生长无影响。高浓度用药组可抑制卵巢癌细胞生长,电子显微镜可观察到细胞凋亡的形态学变化,1×10-6~1×10-4mol／L的TAM和MPA联合用药组在抑制细胞增殖的同时尚可诱导细胞凋亡,其细胞凋亡率分别为4.06％、11.77％、19.3％,两两比较,差异均有显著性(P<0.05)。结论:TAM和MPA联合用药既对顺铂耐药的卵巢癌细胞有杀灭作用,又因其较小的副作用及肯定的临床意义有可能成为卵巢癌有效的化疗辅助用药。更多还原

【Abstract】 Objective: To investigate the effect of tamoxifen combined with medroxyprogesterone on the growth of cisplatin -resistant human epithelial ovarian cancer cell line SKOV - 3/DDP. Methods: SKOV - 3/DDP cells were treated with different concentrations (1× 10-10-1×10-4mol/L) of tamoxifen combined with medroxyprogesterone. . The inhibition of SKOV-3/DDP cell growth was measured by MTT. The morphological change of cell was observed by electron microscopy. Cell cycle and apoptotic percentage were accounted by flow cytometry. Results: Low concentration (1×10-10 -1×10-7mol/L) of tamoxifen combined with medroxyprogesterone had no effect on SKOV-3/DDP cell growth. SKOV -3/DDP cell growth was inhibited by drugs at high concentration (1×10-6-1×10-4mol/L) in a dose-dependent manner (P<0. 01) . With the high concentration (1×10-6-1×10-4mol/L) of drugs, GO/G1 arrest of SKOV-3/DDP cell was enhanced and the apoptotic peak and percentage was increased. Conclusion: Tamoxifen and medroxyprogesterone with the inhibitory effect on the growth of cisplatin-resistant ovarian cancer cells and the minor ill effect could be used to ovarian cancer patients as an adjuvant therapy.更多还原
