庆大霉素抗菌骨制备及体内外释放特性实验研究
Preparation of gentamicin compound bone and sustained gentamicin release in vitro and in vivo
【摘要】 目的研制一种既有成骨作用又有局部抗感染能力,且有较低免疫原性的新型高效骨植入材料。方法采用超声和负压双重复合法制备复合庆大霉素抗菌骨并辐照灭菌,通过扫描电镜和微生物测定法分别检测其复合情况及庆大霉素在体内外的缓释特性。结果明胶复合庆大霉素抗菌骨在体外12d、体内14d持续高于金黄色葡萄球菌最小抑菌浓度,基本达到临床局部抗菌效果。结论明胶复合庆大霉素抗菌骨具有良好的体内外缓释特性,有望成为感染性骨缺损的理想植入材料。
【Abstract】 Objective To explore a new kind of bone graft (gentamicin compound bone), which can have a local anti infective function, low immunogenicity and high osteogenesis. Methods The gentamicin compound bones were prepared by means of ultrasonic and vacuum. After irradiation sterilization, the amount of gentamicin loaded in bone was observed by Scanning Electron Microscope(SEM) and the release of gentamicin in vivo and vitro was measured by microbial determination. Results The concentration of gentamicin exceeding the minimum inhibitory concentration(MIC) of S. aureus lasted for 12 days in vivo, and 14 days in vivo, which can satisfy the clinical requirement of local anti infection. Conclusions The gentamicin compound irradiation bone is of a good sustained release featurein vivo and vitro. It might be an ideal bone grafting material for bone defects with infection.
