脑力智宝对乳鼠脑皮质神经细胞和PC12细胞生长发育的影响

【摘要】 目的:从细胞学角度研究脑力智宝在维持神经元存活和促进神经突起生长方面的保护作用。方法:将SD大鼠分为脑力智宝提取物高剂量组(0.63g/L)、低剂量组(0.04g/L)和对照组,连续给药7次后眼球取血制备含药血清,用MTT法检测脑力智宝对原代培养神经细胞生长发育的影响;用分散培养法培养PC12细胞,测定加入不同浓度脑力智宝提取物(0.63和0.04g/L)、脑力智宝含药血清(6和3g/kg)、神经生长因子后PC12细胞在24,48和72h的突起阳性细胞百分比。结果:脑力智宝对PC12细胞具有营养作用,使细胞突起增加增长,阳性细胞数增多,48h时0.63g/L脑力智宝提取物可使阳性细胞百分数从模型组的17.2%提高到38.4%(t=4.89,P<0.01),3g/kg脑力智宝含药血清可使阳性细胞百分数达到69.0%(t=9.37,P<0.01),并可促进原代培养鼠脑皮质神经细胞的生长发育,MTT法检测A值明显增加,尤以0.63g/L脑力智宝提取物(t=4.79,P<0.01)和3g/kg脑力智宝(P<0.01,t=8.81)组作用明显。0.63g/L脑力智宝提取物可显著促进PC12细胞增殖,在2,4和6d时分别可使PC12细胞数目较对照组分别增加1.56,1.85和2.96倍,差异均有显著性意义(t=2.67,5.27,3.12;P<0.05~0.01),均与对照组比较有显著性差异;6g/kg含药血清组亦显著增加。脑力智宝提取物和含药血清均可促进无血清培养神经?

【Abstract】 AIM:To study the protective effect of nao li zhi bao(NLZB) in maintaining the survival of neuron and improving the outgrowth of neurite by cytology. METHODS: The SD rats were divided into 3 groups: NLZB extract high dosage group (0.63 g/L), low dosage group (0.04 g/L), and control group. After successive administration for 7 times, the drug serum was prepared by drawing blood from eyeball. The effect of NLZB on the growth and development of primary cultured neurons was detected with the MTT.PC12 cells were cultured with the method of dispersed cell culture, and then the positive percentage of PC12 cells neurite outgrowth was determined at 24, 48 and 72 hours after different concentrations of NLZB extract (0.63 and 0.04 g/L) and drug contained serum(6 and 3 g/kg) were added. RESULTS: NLZB had the effect of nutrition on PC12 cells, it could increase the cell process and the number of positive cells. At 48 hours, the positive percentage in the model group was increased from 17.2%to 38.4%by 0.63 g/L NLZB extract (t=4.89, P< 0.01), and it reached 69.0%by 3 g/kg NLZB drug contained serum(t=9.37, P< 0.01), and the growth and development of primarily cultured brain cortical nerve cells could promoted, and the A value detected by MTT was obviously increased,especially in the 0.63 g/L NLZB extract group (t=4.79, P< 0.01) and 3 g/kg NLZB drug contained serum group (t=8.81,P< 0.01). The 0.63 g/L NLZB extract could significantly promote the proliferation of PC12 cells, the PC12 cell numbers at 2, 4 and 6 days were significantly increased by 1.56, 1.85 and 2.96 times as compared with those in the control group (t=2.67, 5.27, 3.12, P< 0.05 to 0.01); and those in the 6 g/kg NLZB drug contained serum group were also significantly increased. Both NLZB extract and drug contained serum could promote the growth of serum free cultured neurite outgrowth, and had the effect in improving growth of primarily cultured nerve cells and could increase the number of positive cell process. CONCLUSION: NLZB has the effect in accelerating the growth and development of brain cortical nerve cells and PC12 cells in neonatal rats.
