复方鳖甲软肝方对活化肝星形细胞细胞因子分泌的影响
Effect of fufang biejia ruangan fang on the secretion of activating hepatic stellate cell cytokine
【摘要】 目的:观察复方鳖甲软肝方药物血清对离体肝星形细胞(hepaticstellatecell,HSC)活化时分泌转化生长因子-β1(transforminggrowthfactor-β1,TGF-β1)和血小板源性生长因子(plateletderivedgrowthfactor,PDGF)的影响,以探讨其抗肝纤维化作用机制。方法:将成年健康雄性SD大鼠75只,随机分为对照组、模型组、高、中、低剂量药物组共5组,并制备各组血清。采用SD大鼠肝星形细胞分离、培养、药物血清制备,免疫组化检测各实验组血清对肝星形细胞分泌TGF-β1,PDGF的影响,并经图像分析系统作定量分析。结果:培养48h模型组TGF-β1,PDGF阳性反应物A值分别为733.58±189.73和659.45±76.56;72h分别为1509.87±77.50和1208.02±132.21,与对照组(48h:570.78±57.61和479.65±38.49;72h:1279.30±147.78和805.30±121.04)比较,差异显著有显著性意义(F=5.833~21.250,P<0.05)。培养48,72h,高、中剂量药物组与模型组TGF-β1,PDGF表达比较,差异也有显著性意义(F=5.833~21.250,P<0.05),可显著抑制TGF-β1,PDGF表达,并呈量效关系。结论:复方鳖甲软肝方药物血清能有效抑制肝纤维化时活化肝星形细胞分泌TGF-β1和PDGF,从而达到抑制肝星形细胞增殖、活化的抗肝纤维化防治作用。
【Abstract】 AIM:To observe the influence on the expression of transforming growth factor beta1(TGF β1) and platelet derived growth factor(PDGF) of hepatic stellate cell(HSC) in vitro by fufang biejia ruangan fang drug serum,and discuss its mechanism of anti hepatic fibrosis. METHODS:Seventy five adult healthy male SD rats were randomly divided into normal control group,model group, high,middle and low dose groups (fufang biejia ruangan fang),and the serum was prepared in all the groups. Different drug serums were added to HSC separated from SD male rat for culture to examine expression of TGF β1 and PDGF of HSC by immunohistochemistry,and analyze quantitatively the outcome with image analysis system(IAS). RESULTS:Expressions of TGF β1 and PDGF positive product of HSC(A value) in the model group were 733.58±189.73 and 659.45±76.56 for cultivating 48 hours,1 509.87±77.50 and 1 208.02±132.21 for 72 hours,which were significantly different from those in the control group(48 hours: 570.78±57.61 and 479.65±38.49; 72 hours: 1 279.30±147.78 and 805.30±121.04)(F=5.833 to 21.250,P< 0.05).After cultivating for 48 and 72 hours,the expressions of TGF β1 and PDGF of HSC in the high and middle dose fufang biejia ruangan fang groups had significant difference from those in the model group (F=5.833 to 21.250,P< 0.05).Fufang biejia ruangan fang drug serum of all doses could significantly inhibit the expressions of TGF β1 and PDGF, and the effect was related to the doses. CONCLUSION:Fufang biejia ruangan fang drug serum effectively inhibits the expressions of TGF β1 and PDGF of HSC.The mechanism of anti hepatic fibrosis by fufang biejia ruangan fang may be related to the reduction of proliferation and activation of HSC by drug.
