川芎嗪对急性癫痫大鼠弓状核神经元损伤的保护作用

【摘要】 目的:研究川芎嗪对急性癫痫大鼠弓状核神经元损伤中的保护作用。方法:30只SD大鼠按随机数字表法随机分为对照组、癫痫组和治疗组,采用光、电镜技术和形态计量方法对弓状核神经元进行形态定量研究。结果:①3组间神经元胞体的面积分数、面数密度、神经元胞体和胞核的等效直径无显著性变化。②对照组、癫痫组和治疗组大鼠弓状核暗神经元细胞器体积分数比较:线粒体犤(5.82±0.91),(3.22±0.66),(5.88±0.43)%犦(F=6.89,P<0.05)、粗面内质网犤3.66±0.35),(5.72±0.79),(3.84±0.36)%犦(F=25.28,P<0.05)和溶酶体犤(1.82±0.53),(3.68±0.72),(2.28±0.49)%犦(F=24.64,P<0.05)的体积分数差异有显著性意义。经SNK检验显示,癫痫组线粒体体积分数下降(q=9.96,P<0.05),粗面内质网(q=10.72,P<0.05)和溶酶体(q=7.15,P<0.05)的体积分数增加,其余未见明显差异(q≤3.08,P<0.05)。③治疗组和对照组间暗细胞各细胞器的体积分数和面数密度差异无显著性意义(F≤3.54,P>0.05)。结论:川芎嗪对癫痫脑损伤具有保护作用。

【Abstract】 AIM:To study the protection of ligustrazine to arcuate nucleus neuron injury in rats with acute epilepsy. METHODS:Thirty SD rats were divided into the control group,epilepsy group and therapy group with the random number table method.A morphometric study of arcuate nucleus neurons was made by light and electron microscopy technique and morphometric method. RESULTS:①There were no significant differences in the area fraction and numerical density of neuron cell body,the equivalent diameter of neuron cell body and nuclear among the three groups.②In the comparison of volume scores of dark neurons of arcuate nucleus among the control group, epilepsy group and therapy group,there were significant differences in the scores of mitochondria[(5.82±0.91)%,(3.22±0.66)%,(5.88±0.43)%](F=6.89,P < 0.05), rough endoplasmic reticulum(RER)[3.66±0.35)%,(5.72±0.79)%,(3.84±0.36)%](F=25.28,P< 0.05), and lysosoms[(1.82±0.53)%,(3.68±0.72)%,(2.28±0.49)%](F=24.64,P< 0.05).The SNK test showed that the volume score of mitochondria decreased(q=9.96,P< 0.05), while those of RER(q=10.72,P< 0.05)and lysosomes(q=7.15,P< 0.05) increased in the the epilepsy group, and there were no obvious differences in others(q≤3.08,P< 0.05).③There were no significant differences in the volume scores and numerical density of the organells of dark cells between the therapy group and the control group(F≤3.54,P >0.05). CONCLUSION:Ligustrazine has an protective effect on the brain injury in rats with acute epilepsy.
