高浓度氧诱导增殖性视网膜病变动物模型的构建及判断

【摘要】 【目的】探讨一种经济实用的增殖性视网膜病变动物模型的构建及判断方法。【方法】将鼠龄为 2d的SD幼鼠 2 0只随机分为对照组和实验组 ,实验组动物置于 80 %高浓度氧环境中饲养 1 0d后转至自然空气中继续饲养 ,而对照组动物则一直置于自然空气中饲养。在鼠龄 2 0d时无痛处死幼鼠 ,取眼球作病理切片 ,计数其视网膜新生血管芽细胞核 ,比较分析其结果。【结果】实验组视网膜新生血管芽细胞核数目达到 (1 91 1± 1 5 7)个 ,远远高于对照组的 (37 2± 2 3 5 )个 ,二者存在着非常显著的差别 (P <0 0 1 )。【结论】高浓度氧能短时间内有效诱导SD新生幼鼠视网膜新血管芽的生长 ,成功构建增殖性视网膜病变动物模型 ;病理组织切片是一个经济有效的判断增殖性视网膜病变构建成功与否的方法。

【Abstract】 To explore economical and useful measures to build and evaluate proliferative retinopathy model.Twenty postnatal 2 days SD newborn rats were randomized to experimental group and control group.The animals of experimental group were raised in hyperoxic enviroment for ten days and then to room air for seven days as well as that of control group were in room air all the time.The eyeballs were enucleated and fixed by formaldehyde at postnatal 20 days and then cross sectioned and stained with hematoxylin and eosin(HE).The nuclei of proliferative retinal vessels were counted through the cross sections.The average nuclei of the new vessels of experimental group were statistically higher than that of control group(191 1±15 7) vs (37 2±23 5).[Conclusion]Hyperoxia can induce neovascularization successfully in newborn rats and lead to proliferative retinopathy.Pathological cross sections are also effective and economical assessment methods for reinal neovascularization and proliferative retinopathy.
