间接ELISA检测减蛋综合征病毒抗体方法的建立

【摘要】 应用SephdexG-200亲和层析获得纯化的减蛋综合征病毒EDSV作ELISA抗原,应用抗鸡Ig轻链单抗1B7辣根过氧化物酶结合物建立了定量检测EDSV抗体水平的ELISA方法。确立了将鸡血清100倍稀释监测ELISA效价(ET)的回归方程y=0.59287x+0.75757(r=0.96763)可用于定量测定。血凝抑制(HI)抗体与ELISA方法的比较试验表明,ELISA法比血凝抑制试验敏感2倍以上.

【Abstract】 The indirect ELISA method for detecting antibody level to EDS-76 virus were developed by preparing coating antigen,purified by Affinity Chromatography using Sephdex G-200 , and enzyme conjugate of mouse against light chain of chicken Ig . A detecting titer of coefficient relation ,y=0.592 87x+0.757 57(r=0.96 763)were developed under 100 fold dilution sera sample. The ELISA titer is 2 times higher than hemagglutination inhibitor test (HI) result.
